MARKERS OF UROTHELIAL CELL PHENOTYPE A prerequisite for studies of epithelial cell regulation is the availability of reliable markers of cell phenotype, In the urothelium, a number of differentiation-associated anti- Urothelial Tissue Regulation 21 gens have been identified. The best characterized moieties are the four major integral membrane proteins, known collectively as uroplakins (UP), which are species-conserved and constitute the specialized AUM plaques of umbrella cells. 33 and interact respectively with the unrelated 15,000 Mr UPII 34 and 47,000 Mr UPIII 35 proteins.

HERI has been implicated in regenerative growth and proliferative diseases of the human bladder epithelium, however a cognate HER I ligand that can act as a growth factor for normal human urothelial cells has not been identified. We have shown that heparin-binding epidermal growth factor-like growth factor (HB-EGF), an activating HERI ligand, is an autocrine regulator of human urothelial cell growth. This conclusion is based on demonstration of HB-EGF synthesis and secretion by primary culture human urothelial cells, identification of HER I as an activatable HB-EGF receptor on human urothelial cell surfaces, stimulation of human urothelial cell clonal growth by HB-EGF, inhibition ofHB-EGF-stimulated growth by heparin and oflogphase growth by CRM 197, a specific inhibitor ofHB-EGF/HERI interaction, and identification of human urothelium as a site of proHB-EGF synthesis in vivo.

FORMATION OF BLADDER TISSUE The direction which we have followed to engineer bladder tissue involves the use of both synthetic and natural biodegradable materials which act as cell delivery vehicles. Urothelial and muscle cells can be expanded in vitro, seeded onto the polymer scaffold, and allowed to attach and form sheets of cells (Figure 4). The cell-polymer scaffold can then be implanted in vivo. We have performed a series of in vivo urologic associated cellpolymer experiments. 4 Implanted cells oriented themselves spatially along the polymer surfaces.
